Human tumor xenograft models are the mainstay of preclinical proof-of-concept testing of all experimental anti-tumor drugs.  The current mouse xenograft models have been primarily based on immunodeficient mice. However, an increasing amount of evidence suggests that immune responses may contribute to the efficacy of some therapies, such as therapies directed to cancer management.  Unfortunately, due to the nature of current xenograft models, it has been very difficult to study the role of immune responses in treatments.    
   
To address the issue of lacking immuno-response in current xenograft models, Biochain is offering a patented, revolutionary, immu-recovered xenograft model that takes into account the contribution of the immune system to the efficacy of a treatment regime.    

In this system, cancer xenograft was first established in the conventional way. Afterward, mouse T-cells were introduced back to the animal to recover and fine-tune the immu-system of the animal, as shown in the graphics below:

 

Living Tumor xenograft model

Living Tumor xenograft models are developed using human tumor tissues grafted in subrenal capsules of SCID mice. The human tumor tissues are developed into transplantable tumor tissue lines. These tumor tissue lines offer a sustainable tumor source for xenograft models. The model is in many aspects superior to conventional tumor model systems which are based on cultured cancer cell lines.

·  Resemblance to human tumor: At the subrenal capsule graft site, the Living Tumor tissue lines retain the cell heterogeneity of the original human cancer and better retain original drug sensitivities, genotype and phenotype.

Description

Patient tissue will be collected and cut into small pieces. Small tissue samples will be implanted into the subrenal capsule of the mouse. The tissue will grow and be ready for following generations of xenografting. After a few generations of grafting, a Living Tumor TM line will be established.


Tarcine will establish tumor lines by the fresh tumor tissues provided by customers or tissues procured by us under customers' request. It will take 6-10 months to generate a new grafted tumor line successfully. DNA/RNA/Protein lysate/Tissue Sections of the custom made tumor lines can be offered separately.

Conventional mouse xenograft model

Xenograft models are a well-established method for anti-cancer drug efficacy screening. Every clinically approved agent for the treatment of cancer has shown activity in conventional preclinical in vivo models, (Sausville and Burger 2006, PDF version) and mouse xenograft models are the most accessible, efficient, and cost-effective in vivo models for anti-cancer efficacy testing.

 

Nude or SCID mice will be used to establish the cancer model. Tumor xenografts will be established by subcutaneous injection of cells on the flank of each animal. Following inoculation, tumor site will be palpated up to three times weekly until the tumor is established. After establishing sufficient tumor volume, mice will be stratified by tumor size and randomly assigned to each of the study cohorts. Animals will be dosed by route and schedule of your choice.
